چکیده

In an earlier report (1) we compared the responses of hepatic glycolytic enzyme activities of normal rats fed either glucose or fructose for 3 days. The enzymes studied were phosphoglucomutase, glucose 6-phosphatase, phosphoglucose isomerase, and the dehydrogenases of glucose B-phosphate and gluconate g-phosphate. When the diets containing free hexoses were substituted for the stock diet, the activities of isomerase and both dehydrogenases were augmented, but the response to fructose feeding exceeded by far that observed with glucose. Only the fructose diet augmented the phosphatase activity, whereas neither diet affected the mutase activity. The present report concerns the responses to hexose feeding of these hepatic enzymes in the diabetic rat. It is shown that the magnitude of response to either hexose by the diabetic rat does not approach that by the normal rat. Our findings indicate that the liver of the diabetic rat is characterized by a reduced response to the substitution of hexose-containing diets.

Effect of Fructose on Lipogenesis from Lactate and Acetate in Diabetic Liver*

Defective glucose utilization in the liver of the glucose-fed diabetic rat has been stated to be the result of at least two blocks: one, at an early stage in glycolysis, probably hexokinase; the other, in the synthesis of fatty acids from the Z-carbon-like intermediate. This conclusion was based on the following observations: (1) the conversion of C14-glucose to CO2 was depressed in the liver o...

Effects of dietary fructose or glucose on triglyceride production and lipogenic enzyme activities in the liver of Wistar fatty rats, an animal model of NIDDM.

Effects of dietary carbohydrates on triglyceride production and hepatic lipogenic enzyme activities were examined in Wistar fatty rats, an animal model of noninsulin dependent diabetes mellitus, fed fructose or glucose and were compared with those of Wistar lean rats. Carbohydrates were supplied in 10% drinking solutions for 21 days. As compared with lean rats, Wistar fatty rats were characteri...

The effect of fructose feeding on glycolytic enzyme activities of the normal rat liver.

A number of studies have brought to light the fact that substitution of fructose for glucose as the sole carbohydrate in a diet results in a profound change in the metabolic behavior of an animal. Thus, the ability of the liver of a glucose-fed, diabetic rat to convert acetate carbon to fatty acids is considerably impaired (l), but when the glucose of the animal’s diet is replaced by fructose, ...
